Can any damage occur from painkillers

I am suffering from arthritis and because of it I must take medications almost constantly. The most used medications are, off course, painkillers. My wife keeps telling me that it is not safe to use painkillers that much. I disagree. Is she or am I right? Can any damage occur from painkillers?
