Hormones and neurotransmitters are released during an orgasm, which is when one ejaculates. In males there is a refactory period (or a cool-down period) in which the nerves of the penis are not responsive to sensation. There are other physical changes in the body that happen after orgasm which take time to be restored. An example of this is the neurotransmitters that are used to maintain an erection take time to build up in the neurons again. Also some enzymes released during orgasm will prevent the penis from becoming erect because they will destroy the neurotransmitter that does so. As for the loss of interest in sexual drive after an orgasm, that is a reaction in the brain caused by the release of endorphines, neurotransmitters and hormones. One hormone is Ocytocin which causes the euphoria (calmness) and cuddling sensation after climax. As with any other drive in the body, such as hunger, thirsts or urination, it is followed by a reward of a pleasurable feeling, followed by a relaxing of that drive to communicate a need. The sex drive is the same and is switched off in the brain after it is satisfied.
