Can someone explain me here why does levodopa become less effective over time? Most of the Parkinson’s patients are treated with this drug. What happens to them with time?

From what I read as Parkinson's disease progresses the number of dopamine cells in the brain continues to decrease. The brain has fewer cells that can take up and store the drug for later release. In these circumstances the brain is said to have lost its 'buffering' capacity and As a result of this loss changes in the levels of the drug in the blood with standard levodopa formulations can no longer be compensated for and these variations are associated with variations in the availability of dopamine in the brain.
