It's importanto define what you mean when you say 'virgin'.
If by virgin, you mean never had sex of any kind with a man, including oral or anal, then no; you cannot tell. Furthermore, even if virgin refers only to a female who's never had a penis inside her vagina, it will still be impossible to verify using physical means because she may have never even been with a man but she may have been penetrated (even by herself) with another object.
However, if virgin means her vagina has never been penetrated (not counting a finger or a tampon, but by a penis-sized object), it is sometimes possible to tell if a girl IS a virgin but not always possible to tell if she is not.
All girls are born with an anatomical structure called the hymen located in the introitus, or entrance to the vagina. Just like the physical appearance of women's genitalia differs, so does the physical characteristics of the hymen. In other words, the don't all look the same.
The hymen is a membrane just inside the entrance to the vagina. It can vary both in shape, size, and most importantly, strength, but it is typically ring-shaped with a small, round, smooth opening in the center. It is usually large enough to pass a tampon or small finger through, but penetration by any significant-sized object (depending on the size and age of the girl) will 'break' the hymen. Make no mistake; she still has a hymen - EVERY girl does - but now the center opening is larger and is no longer smooth.
If the girl was very young, had a particularly stronger or thicker than normal hymen, or was penetrated by a very large or jagged object, there may be physical signs of trauma to the hymen such as a visible tear.
As stated earlier, a girl with a normal or thin hymen, who engages in frequent horseback riding or other activities in which there is REPETITIVE minor 'trauma' to her genitalia, may cause her hymen to gradually weaken or lose it's 'virginal' size or appearance. The vagina of this girl, even if she is a true virgin may be indistinguishable from that of a girl who has previously had intercourse. Interestingly, that's why in cultures where premarital sex is taboo and great value is placed on a girl's virginity, these activities are strongly discouraged.
As far as bleeding:
If a girl tells you she is a virgin and does not bleed after intercourse, it does not necessarily mean she lied. This may have indeed been her first time. She may have had a thinner or larger hymen (you may also have a tiny penis), or her hymen may have been gradually 'weakened' by aforementioned activities.
If a girl tells you she's a virgin (whatever she means by that) and she bleeds after intercourse, all this means is that this was the first time she was vaginally penetrated by a penis or object of similar size. She may have blown half the high school football team and/or been anally raped y by her step father every Sunday since she was 13. You're just the first person to the 'Promised Land'.
So to sum up, if you inspect your girlfriend/fiance/wife (WHICH I DO NOT RECOMMEND) and you do not see her hymen, it does noy necesarily mean she is not a virgin. She in fact be as pure as a nun. If you do find her hymen intact, then she had never had a penis inside her vagina. She may or may not be a virgin depending on what she had and where she had it inside her, and depending on your definition of 'virgin'.

You really need to be more specific with what you mean by 'doing sex'. If a penis has penetrated her vagina, then technically she is not a virgin. Conversely, if a penis has not penetrated her vagina, then she is technically still a virgin.
However whether she is considered a virgin morally or culturally is another matter. Can you give more details of what happened, how it happened, and when it happened. Where is she from?
