Yes when playing with the genitals or masturbating there is increased blood flow which leads to bigger more noticeable veins. The glans also becomes more sensitive temporarily but over time masturbation does not lead to hypersensitivity of the glans if anything it may slightly decrease sensitivity especially if you are uncircumcised
