Anything goes into the vagina you have lost your virginity. Now, I would personally say (and others may disagree which is fine with me) that you lose your virginity when you have a penis inserted into you. Everyone will say something different.

If it's in the "spiritual" sense, then you only really lose it by having sex.

I know in many cultures they want to marry virgins but realistically and biologically, there is no way to tell if someone is a virgin or not, so it's all a bit silly really.
