It sounds like you really DO care whether or not they're gay and it also sounds like they are very aware that you care if their first response when questioned was that they are not gay.
All children are curious, boys especially, and want to compare what they have to what the other kids has. It almost always means nothing. Every kid (yes, every kid) has had some experience of this type and it's completely normal and nothing to get worried about. The only real damage would come from you over-reacting about it.
